ProLISA HCV 3rd Gen is an in-vitro enzyme immunoassay for the qualitative determination of hepatitis C antigen in human serum or plasma. It is intended for screening of blood donors or other individuals at risk forhepatitis C infection and for clinical diagnostic testing.

Overview:

  • HCV 3.0 ELISA is an in vitro enzyme immunosorbent assay for qualitative determination of Hepatitis C in human serum or plasma.

  • Primarily affects the liver.

  • HCV virus is transmitted through:

  • Sexual contact

  • Contaminated blood and blood products

  • Blood transfusion


 

Principle:

“Indirect ELISA technique.”

  • Recombinants of HCV antigens(CORE+NS3+NS4+NS5) are coated onto microwells.

  • Samples with positive and negative controls are added in the coated wells and incubated.

  • Wash the wells to remove unbound components and anti human IgG HRP conjugate is added.

  • After washing wells to remove unbound enzyme, substrate is added.

  • Stop solution is added

  • Color is read on photometer at 450/650 nm wavelength.



 

Procedure:

  1. Add 100µl of sample diluent in each well

  2. Add 10µl of control or specimen in separate wells

  3. Cover the plate with aluminum foil and incubate for 30mins at 37°C

  4. Wash each well 6 times with 300µl of wash buffer and dry with absorbent paper

  5. Add 100µl of conjugate in each well and incubate at 37°C for 30 mins

  6. Wash each well 6 times with 300µl of wash buffer and dry with absorbent paper

  7. Add 100µl of substrate solution in each well and incubate at RT for 30mins

  8. Add 100µl of stop solution to stop reaction

  9. Read results at 450/620nm


 

  • Store all kit components at 28°C when not in use.

  • Sensitivity 100% Specificity >=98%
